Fixation of biological specimens

چکیده

Fixation is usually the first step to prepare biological specimens for microscopy. Any treatment which will preserve cell structure and its biochemical composition can be deemed to be fixation. Of course, the quality of fixation is the key for all following steps which are necessary in histological research. Hence, preservation of cells with minimal alteration of morphology and virtually no loss of molecules is essential in tissue preservation. In this sense, tissue fixation has to preserve the cells in as life-like a state as possible. Moreover, fixation should protect biological specimens from the denaturing effects of dehydration and all further processings.

A New Method of Brain Plastination

Introduction: Plastination is a unique technique for preservation of biological specimens used for teaching purposes. The protocol of flexible sheet plastination includes fixation, slicing, dehydration, force impregnation, casting, and curing (Hajian, Rabiei, Fatollahpour & Esfandiary, 2008). The procedure is done by using P87 flexible unsaturated polyester resin and provides heavy, gross, frag...

P96 Technique for Tissue Plastination in Preparing Long-Lasting Anatomical Specimens

Background: Plastination is a technique of long-term preservation of the natural tissues without the typical problems related to wet specimens. Plastinated specimens are durable, dry, and flexible. In this process, the water and fat in biological tissues are substituted by a curable polymeric resin, i.e., subsequently cured. In this study, a new unsaturated polyester resin (P96) was synthesized...

Fixation of biological specimens by high hydrostatic pressure.

We investigated whether or not high hydrostatic pressure could be used as a new method of histological fixation. Various high hydrostatic pressures from 200 to 800 MPa (Mega Pascal) were applied to fresh organs including heart, liver, intestine and kidney of mice, and the gross and light microscopic findings were compared with those of organs fixed with formalin and those without fixation. The ...

Evaluation of the difference in surgical margin values in excisional skin lesions during surgery and after formalin fixation

Background: Surgical margin determination in malignant lesions is essential and has a direct impact on the choice of postoperative treatments and patient follow-up. Therefore, the processes affecting this variable are worth exploring. This study aimed to evaluate the difference in surgical margin values in excisional skin samples during surgery and after formalin fixation. Methods: A cross-sec...

A Novel Fixation Method for Variable-Sized Endoscopic Submucosal Dissection Specimens: An In Vitro Animal Experiment

BACKGROUND Endoscopic submucosal dissection is considered a curative and minimally invasive treatment for early gastric cancer; however, precise pathologic assessment of resected specimens is required to develop further treatment plans. Human error during specimen handling can affect objective assessment of resected specimens. In this study, we investigated whether a novel tissue fixation devic...
